Callogenesis and Micropropagation of Grape (Vitis Vinifera L.) (Paperback)


The present study deals with the callogenesis and micropropagation of grapes (Vitis vinifera L.) and biochemical investigation of protein content and peroxidase activity during different stages of growth. In the protocol of micropropagation, four different types of media with different concentration of auxin, cytokinin and their combinations were used. Best result was obtained with the TDZ 25.0 (uM) with 90% frequency rate. In the induction of callogenesis procedure, three different types of media concentration of auxin, cytokinin and their combination were used with leaf and node as explant. The best result observed from leaf explant obtained from 2,4-D 3.0 (mg/l) with maximum 90% frequency rate. Best result for callus formation from nodal explant obtained from 2,4-D 3.0 + BAP 1.0 + 1.0 TDZ (mg/l) with 90% frequency rate.In the estimation of soluble protein content best result of in vivo grown cultures obtained from leaf explant (0.085 0.643) in 3rd subculture. In the comparison of specific activity of peroxidase enzyme, best result of in vivo grow cultures obtained from shoot tip explant (0.00063 0.692) in 3rd subculture.
